以四氫吡喃-4-羧酸甲酯為原料合成四氫吡喃-4-甲酰胺的一般步驟如下:將四氫-2H-吡喃-4-羧酸甲酯(7g,48.6mmol)與30%氨水(20mL)混合于密閉反應(yīng)瓶中,室溫下攪拌反應(yīng)18小時(shí)。反應(yīng)完成后,通過(guò)減壓蒸餾除去過(guò)量的氨,殘余物用乙醇進(jìn)行重結(jié)晶,最終得到5.6g(產(chǎn)率89%)目標(biāo)產(chǎn)物四氫-2H-吡喃-4-甲酰胺。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(401MHz,DMSO-d6)確認(rèn),化學(xué)位移δppm分別為7.21(br.s,1H),6.73(br.s,1H),3.90-3.80(m,2H),3.30-3.23(m,2H),2.36-2.24(m,1H),1.66-1.47(m,4H)。
