以化合物(CAS: 188290-36-0)為原料合成2,5-二溴噻吩的一般步驟如下:參照文獻方法(Kellogg RM, Schaap AP, Harper ET, Wynberg NDH. Acid-catalyzed bromination, deuteration, rearrangement, and debromination of thiophenes under mild conditions. J Org Chem 1968; 33: 2902-9)進行制備。向噻吩(14.0 g, 0.16 mol)溶于250 mL 50:50(v/v)乙酸和氯仿混合溶劑的溶液中,加入N-溴代琥珀酰亞胺(60 g, 0.33 mol)。將反應混合物回流15分鐘。反應完成后,冷卻至室溫,加入水,用氯仿進行萃取。合并有機相,用無水硫酸鈉干燥,減壓濃縮除去溶劑。粗產(chǎn)物通過柱色譜法純化,以己烷為洗脫劑,得到目標化合物2,5-二溴噻吩,為液體(31.5 g, 產(chǎn)率84%)。
