制備方法

通用方法:將四氫噻喃-4-酮(40.10 g,0.271 mol)溶于甲醇(700 mL)中,冰浴冷卻下分批加入硼氫化鈉(15.36 g,0.406 mol),攪拌反應(yīng)1小時(shí)(注意:反應(yīng)放熱劇烈)。隨后,將反應(yīng)混合物于20°C下繼續(xù)攪拌2小時(shí)。反應(yīng)完成后,減壓濃縮除去溶劑,得到固體殘余物。向殘余物中加入飽和碳酸氫鈉水溶液(150 mL)和乙酸乙酯(250 mL)的混合液(注意:產(chǎn)物易溶于水)。劇烈振蕩后,分離有機(jī)層,水層用乙酸乙酯(4×250 mL)進(jìn)一步萃取。合并所有有機(jī)層,用無(wú)水硫酸鈉干燥,減壓濃縮至干,得到目標(biāo)產(chǎn)物四氫-2H-硫代吡喃-4-醇。
