以4-羥基-6-甲氧基喹啉-3-羧酸乙酯為原料合成4-羥基-6-甲氧基喹啉-3-羧酸的一般步驟:將4-羥基-6-甲氧基喹啉-3-羧酸乙酯(17.8 g,72 mmol)懸浮于170 mL 2 N氫氧化鈉水溶液中,加熱至90℃,維持反應(yīng)1.5小時。反應(yīng)完成后,將混合物冷卻至室溫,緩慢倒入冰水中,并用2 N鹽酸調(diào)節(jié)pH至5。析出的固體經(jīng)過濾收集,用冷水洗滌,干燥后得到4-羥基-6-甲氧基喹啉-3-羧酸(15.8 g,72 mmol,收率99%)。產(chǎn)物經(jīng)1H-NMR(DMSO-d6)表征:δ= 3.91(s,3H),7.53(dd,1H),7.64(d,1H),7.82(d,1H),8.80(d,1H),13.63(bs,1H),15.55(s,1H)。LC-MS分析結(jié)果:保留時間(Rt)= 1.20分鐘;質(zhì)譜(MS):m/z = 220 [M+H]+。
