在氮?dú)獗Wo(hù)下,將(S)-N-芐氧基羰基-L-色氨酸(2.45 g, 7.2 mmol)懸浮于10 mL無水二氯甲烷中,加入4-二甲氨基吡啶(DMAP, 0.1 g, 0.82 mmol)和甲醇(0.28 g, 11.8 mmol)。將反應(yīng)混合物冷卻至0℃,緩慢滴加溶解于二氯甲烷的N,N'-二環(huán)己基碳二亞胺(DCC)溶液。滴加完畢后,將反應(yīng)體系逐漸升溫至室溫并持續(xù)攪拌12小時。反應(yīng)完成后,通過過濾去除不溶物,濾液經(jīng)減壓濃縮。粗產(chǎn)物通過快速柱色譜法(洗脫劑:正己烷/乙酸乙酯,4:1至2:1梯度洗脫)純化,得到目標(biāo)化合物(S)-2-(((芐氧基)羰基)氨基)-3-(1H-吲哚-3-基)丙酸甲酯(2.5 g, 98%)為無色油狀物。薄層色譜(TLC)Rf = 0.19(展開劑:乙酸乙酯/正己烷=1:2)。核磁共振氫譜(1H NMR, 300 MHz, CDCl3):δ 8.94(寬峰, 1H), 7.49(d, J = 7.7 Hz, 1H), 7.25-6.98(多重峰, 9H), 6.81(單峰, 1H), 5.84(d, J = 8.7 Hz, 1H), 4.98(d, J = 3.2 Hz, 2H), 4.68(四重峰, J = 7.3 Hz, 1H), 3.25(單峰, 3H), 3.21(多重峰, 2H)。核磁共振碳譜(13C NMR, 75 MHz, CDCl3):δ 172.2, 155.9, 141.8, 136.8, 134.9, 128.6, 127.8, 126.9, 123.4, 122.3, 120.0, 118.4, 112.1, 109.4, 67.0, 65.1, 55.1。傅里葉變換紅外光譜(FTIR, KBr, 純樣):ν 1707, 1508, 1456, 1436, 1215, 742 cm-1。高分辨質(zhì)譜(HRMS, EI):計算值C20H20O4N2 [M]+ 352.1418,實(shí)測值352.1425。
