在火焰干燥的圓底燒瓶(RBF)中,將氫化鋁鋰(LAH,45.2 mL,45.2 mmol,1M于四氫呋喃(THF)中)的懸浮液冷卻至0°C。隨后,緩慢滴加1H-吡唑-4-羧酸乙酯(3.17 g,22.6 mmol)溶解于THF(20 mL)中的溶液。反應(yīng)混合物逐漸升溫至室溫,并在室溫下攪拌過(guò)夜。反應(yīng)完成后,將混合物置于冰浴中冷卻,并依次逐滴加入1.36 mL水和10 mL 1M氫氧化鈉(NaOH)溶液以小心淬滅反應(yīng),隨后攪拌20分鐘。加入固體硫酸鎂(MgSO4)以干燥反應(yīng)混合物,移除冰浴后,在室溫下繼續(xù)攪拌30分鐘。通過(guò)硅藻土(CELITE)過(guò)濾去除固體,濾餅分別用THF和甲醇(MeOH)洗滌。合并濾液并蒸發(fā),得到目標(biāo)產(chǎn)物(1H-吡唑-4-基)甲醇(1.75 g,產(chǎn)率78.9%),為白色固體。產(chǎn)物經(jīng)核磁共振氫譜(1H NMR,500 MHz,DMSO-d6)確認(rèn):δ 12.58(寬單峰,1H),7.58(單峰,1H),7.40(單峰,1H),4.74(三重峰,J = 5.5 Hz,1H),4.37(雙峰,J = 5.2 Hz,2H)。
