一般步驟:將1,4-二氮雜雙環(huán)[2.2.2]辛烷(DABCO,1.0摩爾當(dāng)量)加入到(2R,3S,4S,5R,6S)-2-(羥甲基)-6-(苯硫基)四氫-2H-吡喃-3,4,5-三醇(10.0g,51.5mmol)在乙酸酐(1.2摩爾當(dāng)量/羥基基團(tuán))中的懸浮液中,室溫下攪拌混合物。反應(yīng)進(jìn)程通過(guò)薄層色譜(TLC)監(jiān)測(cè),洗脫液為乙酸乙酯:正己烷(2:3)。反應(yīng)完成后,將混合物倒入碎冰中,攪拌數(shù)分鐘,促使(2R,3R,4S,5R,6R)-2-(乙酰氧基甲基)-6-(苯硫基)四氫-2H-吡喃-3,4,5-三基三乙酸酯沉淀析出,同時(shí)DABCO溶于水(通過(guò)TLC和碘蒸氣顯色確認(rèn))。產(chǎn)物通過(guò)抽濾分離,隨后用冰水洗滌并干燥。所得產(chǎn)物純度足夠高,可直接用于后續(xù)反應(yīng),產(chǎn)量為18.3g,收率98%。分析純樣品可通過(guò)95%乙醇重結(jié)晶獲得。需注意,加入DABCO及攪拌過(guò)程中反應(yīng)混合物溫度會(huì)升高。在10-20g規(guī)模的實(shí)驗(yàn)中,這一現(xiàn)象有利于顯著提高反應(yīng)速率。然而,對(duì)于更大規(guī)模(50-100g及以上)的反應(yīng),建議分批加入DABCO以避免反應(yīng)混合物過(guò)熱。
