2.1 鹽酸2-萘基肼的合成 在2-萘胺(800 mg,5.59 mmol)的鹽酸(6.0 M,6 mL)懸浮液中,于水-冰浴冷卻條件下,緩慢加入亞硝酸鈉(NaNO2,578 mg,8.38 mmol)的水(1.2 mL)溶液(加入時(shí)間為2分鐘)。將反應(yīng)混合物在水-冰浴中攪拌1小時(shí),隨后緩慢加入氯化亞錫(SnCl2,3.71 g,19.56 mmol)(加入時(shí)間為5分鐘)。將所得懸浮液繼續(xù)在水-冰浴中攪拌3.5小時(shí),之后進(jìn)行過濾。濾餅依次用冰水(0°C,4×8 mL)、室溫水(1×8 mL)、冰乙醚(0°C,2×4 mL)以及乙醚/己烷(1:1,2×4 mL)和己烷(2×5 mL)洗滌。固體經(jīng)干燥后,得到690 mg目標(biāo)產(chǎn)物(Rf = 0.7,40%乙酸乙酯/己烷展開劑,乳白色固體,產(chǎn)率63%)。 1H NMR (DMSO-d6, 250 MHz, δ): 7.81 (m, 2H, ArH), 7.71 (d, J = 7.7 Hz, 1H, ArH), 7.49-7.19 (m, 4H, ArH). MS-El+ m/z: 159.1 (M-HCl+1).
