以2-氰基吡嗪為原料合成2-氨甲基吡嗪的一般步驟如下:參照文獻(JP A 2001-894594)所述方法,具體操作如下。原料2-氰基吡啶購自Sigma-Aldrich Co.LLC。將1.05g(10mmol)的2-氰基吡啶和100mg的60wt% Ni/SiO2催化劑置于20mL甲苯中,裝入高壓釜(型號5U5316)內(nèi),隨后用氬氣置換釜內(nèi)空氣。在氫氣氛圍下加壓至50atm,于140℃反應(yīng)溫度下攪拌反應(yīng)4小時。反應(yīng)完成后,將反應(yīng)液過濾除去催化劑,濾液經(jīng)濃縮處理,定量得到目標(biāo)產(chǎn)物2-氨甲基吡啶(2-AMPZ)。產(chǎn)物經(jīng)1H-NMR(399.78MHz, CDCl3)表征:δ 8.60-8.45(m, 3H), 4.07(s, 2H), 1.79(br, 2H)。
