在0℃下,向2-溴-6-羥基吡啶(0.76 g,4.4 mmol,1.0當(dāng)量)的無水THF(20 mL)溶液中緩慢加入2M異丙基氯化鎂(i-PrMgCl)的THF溶液(2.2 mL,4.4 mmol,1.0當(dāng)量),歷時5分鐘。將澄清溶液在該溫度下繼續(xù)攪拌5分鐘后,于5分鐘內(nèi)逐滴加入2.5M正丁基鋰(n-BuLi)的己烷溶液(3.5 mL,8.8 mmol,2.0當(dāng)量),同時控制反應(yīng)溫度低于20℃。反應(yīng)混合物在相同溫度下攪拌0.5小時,隨后通入干燥的二氧化碳(0.20 g,4.4 mmol,1.0當(dāng)量)。將反應(yīng)體系在0.5小時內(nèi)緩慢升溫至20℃,并用去離子水(6 mL)淬滅反應(yīng)?;旌衔镌?0℃以下攪拌10分鐘,分液后,水相用乙酸乙酯再萃取一次。合并有機(jī)相,通過0.5 cm硅膠短柱過濾,并用乙酸乙酯(10 mL)洗脫。濃縮濾液,殘余物經(jīng)硅膠柱色譜純化(洗脫劑:石油醚/乙酸乙酯=10:1),得到6-羥基吡啶-2-羧酸(產(chǎn)物3m)為灰白色固體(0.56 g,產(chǎn)率93%),熔點(diǎn)為275-277℃。1H NMR (600 MHz, DMSO-d6) δ 7.56 (dd, J = 8.9, 7.0 Hz, 1H), 6.97 (d, J = 6.8 Hz, 1H), 6.65 (d, J = 9.0 Hz, 1H)。13C NMR (151 MHz, DMSO-d6) δ 163.28, 162.67, 140.51, 137.97, 123.88, 110.42。
