以2-異丙基苯胺為原料合成2-異丙基碘苯的一般步驟:將2-異丙基苯胺(27.04 g,0.2 mol)溶解于25%硫酸水溶液(w/w,160 mL)中,在-20℃條件下,緩慢滴加亞硝酸鈉(14 g)的水溶液(26 mL)進(jìn)行重氮化反應(yīng)。將所得重氮鹽溶液轉(zhuǎn)移至碘化鉀(100 g)的水溶液(100 mL)中,反應(yīng)18小時(shí)后,加入氫氧化鈉(36 g)和水(125 mL)以中和反應(yīng)體系。反應(yīng)混合物用己烷萃取,合并有機(jī)相并通過(guò)硅膠柱層析純化,濃縮后得到目標(biāo)產(chǎn)物2-異丙基碘苯,產(chǎn)量35.8 g,收率73%。產(chǎn)物經(jīng)質(zhì)譜鑒定:m/z 246(M+)。1H NMR(CDCl3)δppm:1.23(d,6H,J=6.84 Hz),3.18(septet,1H,J=6.84 Hz),6.86(m,1H),7.28(m,2H),7.82(dd,1H,J=1.24和7.92 Hz)。
