以5-溴-3,4-二氫異喹啉-1(2H)-酮(4.3 g,18.9 mmol)和2,3-二氰基-5,6-二氯-1,4-苯醌(8.6 g,37.9 mmol)為原料,在1,4-二惡烷(76 mL)中混合。將反應(yīng)混合物在100℃下攪拌24小時(shí)。反應(yīng)完成后,蒸發(fā)去除溶劑,將殘余物溶解于乙酸乙酯(500 mL)中,并用10%氫氧化鈉水溶液(2×500 mL)洗滌。分離有機(jī)層和水層,水層進(jìn)一步用乙酸乙酯(4×300 mL)萃取。合并所有有機(jī)層,用無水硫酸鈉干燥,然后蒸發(fā)濃縮。產(chǎn)物通過快速色譜法純化,洗脫劑為二氯甲烷與甲醇的混合溶劑(比例從99:1漸變至96:4),得到5-溴異喹啉-1(2H)-酮(1.49 g,6.65 mmol,收率35%),為黃色固體。產(chǎn)物經(jīng)LCMS分析顯示純度為94%,保留時(shí)間(Rt)為1.243分鐘,電噴霧質(zhì)譜(ESMS)顯示分子離子峰m/z為224([M+H]+)。
