將2,4-二溴丁酸甲酯(10.01 g,38.49 mmol,1.0當(dāng)量)溶于甲醇(15 mL)中,緩慢滴加至0℃下攪拌的三乙胺(11.68 g,16.01 mL,115.5 mmol,3.0當(dāng)量)的甲醇(40 mL)溶液中。滴加完畢后,繼續(xù)攪拌15分鐘。隨后,逐滴加入芐胺(4.33 g,4.41 mL,40.4 mmol,1.05當(dāng)量)的甲醇(25 mL)溶液,滴加完成后繼續(xù)攪拌30分鐘。然后將反應(yīng)混合物緩慢升溫至室溫,并攪拌過(guò)夜。反應(yīng)完成后,將混合物真空濃縮,用二乙醚(100 mL)稀釋,過(guò)濾除去不溶性固體。有機(jī)相用水(200 mL)洗滌,水相再用乙醚(3×50 mL)反萃取。合并有機(jī)相,用無(wú)水硫酸鈉干燥,過(guò)濾后真空濃縮。粗產(chǎn)物通過(guò)柱色譜(25%乙酸乙酯/己烷)純化,得到1-芐基氮雜環(huán)丁烷-2-羧酸甲酯,為橙色油狀物(2.02 g,收率26%)。薄層色譜(TLC)Rf值為0.35(乙酸乙酯:己烷=1:3)。紅外光譜(neat,cm?1): 2953, 2837, 1737, 1442, 1201, 1172。1H NMR(300 MHz, CDCl3)δ: 7.35–7.17(m, 5H, ArH), 3.84–3.70(m, 2H, CH2Ph), 3.62(s, 3H, OCH3), 3.58(d, 1H, J=12.5 Hz, COCH), 3.35–3.27(m, 1H, NCH2a), 2.98–2.88(m, 1H, NCH2b), 2.36(dt, 1H, J=8.7, 17.6 Hz, CHCH2a), 2.25–2.13(m, 1H, CHCH2b)。13C NMR(75 MHz, CDCl3)δ: 173.10, 137.08, 129.17, 128.36, 127.32, 64.40, 62.45, 51.83, 50.85, 21.72。高分辨質(zhì)譜(HRMS, ESI)m/z: 206.1198([M+H]+, 計(jì)算值206.1175)。
