2-溴吡啶與三丁基氯化錫反應(yīng)制備2-三丁基甲錫烷基吡啶的一般步驟:在-78℃下,將丁基鋰(2.5M的己烷溶液,6.33mmol)滴加至新鮮蒸餾的2-溴吡啶(1g,6.33mmol)的四氫呋喃(12mL)溶液中,并進(jìn)行脫氣處理。攪拌30分鐘后,向微紅色溶液中加入三丁基氯化錫(1.7mL,6.33mmol),繼續(xù)在-78℃下攪拌1小時(shí),隨后升至室溫?cái)嚢?小時(shí)。反應(yīng)完成后,用飽和氯化銨溶液淬滅反應(yīng),并用二乙醚萃取。有機(jī)相用飽和氯化鈉溶液洗滌,無(wú)水硫酸鎂干燥,減壓濃縮。殘余物通過硅膠柱色譜(洗脫劑:己烷/乙酸乙酯=20/1)純化,得到目標(biāo)產(chǎn)物2-三丁基甲錫烷基吡啶,產(chǎn)率為94%。1H NMR(CDCl3)δ(ppm):8.73(ddd,J = 4.9,1.9,1.0,1H,H6),7.48(dt,J = 7.4,1.8,1H,H5),7.39(dt,J = 7.4,1.6,1H,H3),7.10(ddd,J = 6.9,4.9,1.7,1H,H4),1.70-1.05(m,18H,CH2),0.85(t,9H,J = 7.3,CH3)。
