在持續(xù)攪拌下,將反應(yīng)體系溫度維持在5-10℃,緩慢滴加氯氧化磷(POCl3,2.75 mL,0.03 mol)至N,N-二甲基甲酰胺(DMF,3.65 mL,0.05 mol)中。隨后,在相同溫度下,向上述冷卻的混合液中緩慢加入溶解于DMF(6 mL)的3-(N,N-二乙基氨基)苯酚(0.01 mmol)。將反應(yīng)混合物在75℃下恒溫?cái)嚢?小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,并緩慢倒入60 mL冰水中。用碳酸鈉溶液中和反應(yīng)混合物,析出棕色固體。通過(guò)過(guò)濾收集固體產(chǎn)物,用冷水洗滌,干燥后從乙醇中重結(jié)晶,得到純品4-(二乙氨基)水楊醛,產(chǎn)率為80%,熔點(diǎn)為62℃(文獻(xiàn)值:62℃)。
