以2-氨基-4-甲基嘧啶為原料合成2-氨基-4-甲基-5-溴嘧啶的一般步驟:向4-甲基嘧啶-2-基胺(10.9 g,100 mmol)的氯仿(400 mL)溶液中加入N-溴代琥珀酰亞胺(17.8 g,100 mmol)。將反應混合物在避光條件下攪拌15小時。反應完成后,將混合物轉移至二氯甲烷(140 mL)中,依次用1N氫氧化鈉溶液(3×200 mL)和飽和氯化鈉溶液(100 mL)洗滌。有機相用無水硫酸鈉干燥,過濾后濃縮,得到5-溴-4-甲基嘧啶-2-基胺(18.8 g,收率99%)。產(chǎn)物經(jīng)LCMS檢測(m/z):188.0/190.0(MH+)。1H NMR(CDCl3)數(shù)據(jù):δ8.22(s,1H),5.02(bs,2H),2.44(s,3H)。
