以2-氯喹啉為原料合成2-肼基喹啉的一般步驟如下:首先將21.0g(128.4mmol)的2-氯喹啉溶解于210mL乙醇中。隨后加入64.3g(1.3mol)水合肼,將反應(yīng)混合物在回流條件下攪拌16小時(shí)。反應(yīng)完成后,將混合物冷卻至0℃,通過過濾收集析出的固體產(chǎn)物,并用少量乙醇洗滌。初步干燥后,將產(chǎn)物置于高真空下進(jìn)一步干燥。最終得到14.5g產(chǎn)物,收率為71%。產(chǎn)物經(jīng)LC-MS(方法1)分析:保留時(shí)間Rt = 1.95min;質(zhì)譜(ESIpos):m/z = 160 [M + H]+;1H-NMR(400MHz,DMSO-d6)數(shù)據(jù)如下:δ= 8.08(寬峰,1H),7.87(雙峰,1H),7.63(雙峰,1H),7.57-7.43(多重峰,2H),7.16(三重峰,1H),6.85(雙峰,1H),4.35(寬峰,2H)。
