以尼爾雌醇雜質(zhì)1和三甲基硅乙炔為原料合成(8R,9S,13S,14S,17R)-3-(環(huán)戊氧基)-17-乙炔基-13-甲基-7,8,9,11,12,13,14,15,16,17-十氫-6H-環(huán)戊并[a]菲-17-醇的一般步驟如下:首先,按照文獻(xiàn)方法從19-羥基雄甾-4-烯-3,17-二酮制備1和1x的混合物,總收率為80%。在惰性氣氛下,將1-混合物(2.04g,6.0mmol)、原甲酸三環(huán)戊酯(8.04g,30mmol)和TfOH(552μL,6.0mmol)在環(huán)戊醇(60mL)中的溶液于100℃攪拌2小時(shí)。反應(yīng)完成后,用飽和NaHCO3溶液淬滅反應(yīng)。減壓除去有機(jī)溶劑,殘余物用乙酸乙酯稀釋,并用鹽水洗滌。有機(jī)相經(jīng)Na2SO4干燥后,減壓濃縮,通過硅膠柱色譜分離得到2b-混合物,為白色固體(1.63g)。接著,在惰性氣氛下,用干燥THF多次洗滌Raney Ni(1.6g)。將2b-混合物(1.63g)的無水THF(180mL)溶液加入Raney Ni中。懸浮液在室溫下攪拌,并通過HPLC監(jiān)測(cè)反應(yīng)進(jìn)程。反應(yīng)完成后(1.5小時(shí)),過濾懸浮液。濾液經(jīng)濃縮后,通過硅膠柱色譜分離得到雌酮3-環(huán)戊基醚(2b),為白色固體(1.62g,總收率79%)。隨后,在惰性氣氛下,于-20℃將n-BuLi(1.2mL,3.0mmol,2.5N)加入乙炔基三甲基硅烷(424μL,3.0mmol)的THF(2mL)溶液中。0.5小時(shí)后,在-20℃下將酮2b(338mg,1.0mmol)的THF(4mL)溶液加入乙炔鋰溶液中。1小時(shí)后,向混合物中加入甲醇(6mL)和KOH(224mg,4mmol),并在室溫下攪拌0.5小時(shí),隨后用2N鹽酸淬滅反應(yīng)。減壓除去有機(jī)溶劑,殘余物用乙酸乙酯稀釋,并用飽和NaHCO3和鹽水洗滌。有機(jī)相經(jīng)Na2SO4干燥后,減壓濃縮,通過硅膠柱色譜分離得到目標(biāo)產(chǎn)物,為白色固體(340mg,93%)。產(chǎn)物的熔點(diǎn)為94-96℃;[α]D23.4 = +11.3(c 1,CHCl3);1H NMR(400MHz,CDCl3)δ7.19(d,J=8.6Hz,1H),6.68(dd,J=8.5,2.3Hz,1H),6.61(d,J=2.7Hz,1H),4.74-4.70(m,1H),2.91-2.79(m,2H),2.61(s,1H),0.89(s,3H);13C NMR(101MHz,CDCl3)δ155.9,137.8,132.0,126.2,115.5,112.9,87.5,79.9,79.0,74.0,49.4,47.1,43.5,39.4,38.9,32.9,32.7,29.8,27.3,26.3,24.0,22.8,12.7;HRMS(ESI)計(jì)算值C25H33O2[M+H]+:365.2402,實(shí)測(cè)值365.2472。
