在圓底燒瓶(RBF,50mL)中,將苯并呋喃(12;1.00g;3.57mmol)溶于DMF(18mL),隨后加入乙硫醇鈉(NaSEt,455mg)。將反應(yīng)混合物加熱至125-130℃,保持1.0小時。反應(yīng)完成后,迅速將混合物倒入兩倍體積的氯化銨(NH4Cl)水溶液中淬滅,并用乙酸乙酯(EtOAc,4×75mL)進(jìn)行萃取。合并有機(jī)相,依次用蒸餾水(H2O)和飽和氯化鈉(NaCl)水溶液洗滌。有機(jī)相經(jīng)無水硫酸鎂(MgSO4)干燥后,過濾并減壓濃縮。粗產(chǎn)物通過硅膠(SiO2)柱色譜純化,洗脫劑比例為4:1的己烷(Hex)與乙酸乙酯(EtOAc),得到目標(biāo)產(chǎn)物2-乙基-3-對羥基苯甲酰基苯并呋喃(13),為白色固體(857mg;3.22mmol;收率90%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(400MHz,CDCl3)、13C-NMR(100MHz,CDCl3)和LC-MS/MS分析確認(rèn)。1H-NMR數(shù)據(jù):δ 10.4(bs,1H;D2O中可交換),7.80-7.78(d,2H),7.49-7.47(d,1H),7.43-7.41(d,1H),7.30-7.26(t,1H),7.22-7.18(t,1H),6.98-6.94(d,2H),2.95-2.89(q,2H),1.36-1.32(t,3H)。13C-NMR數(shù)據(jù):δ 192.1,165.9,161.5,153.6,132.2,130.9,129.0,124.4,123.5,121.1,116.1,115.6,111.0,11.8,12.2。LC-MS/MS數(shù)據(jù):m/z 267.0 → 121.2;GS1和GS2均為20,DP=46,CE=29,CXP=6,tR=4.31min。
