以丙烯酰氯和三(羥甲基)氨基甲烷為原料,按照Pucci等人報(bào)道的方法(Eur. Polym. J. 1991, 27, 1101)合成N-丙烯酰(三羥甲基)氨基甲烷。具體步驟如下:在0℃下,向攪拌的三(羥甲基)氨基甲烷(3.00 g, 24.8 mmol)的3N氫氧化鉀甲醇溶液中,緩慢滴加丙烯酰氯(3.60 mL, 44.6 mmol),保持反應(yīng)體系的pH值在8至8之間。反應(yīng)混合物在0℃下繼續(xù)攪拌3小時(shí),隨后自然升溫至室溫并繼續(xù)反應(yīng)4小時(shí)。反應(yīng)完成后,過(guò)濾混合物,濾液在真空下蒸發(fā)至干。所得固體通過(guò)甲醇重結(jié)晶純化,得到目標(biāo)化合物N-丙烯酰(三羥甲基)氨基甲烷(3.78 g, 87%),為白色粉末,熔點(diǎn)為205℃。產(chǎn)物的表征數(shù)據(jù)如下:IR (NaCl, cm^-1): 3420 (br, s), 1653 (s), 1560 (m), 1540 (m), 1018 (m); 1H NMR (300 MHz, DMSO-d6): δ 3.56 (d, 6H, J = 5.7 Hz, CH2), 4.76 (t, 3H, J = 5.7 Hz, OH), 5.54 (dd, 1H, J = 2.4, 9.9 Hz, Ha), 6.02 (dd, 1H, J = 2.4, 17.1 Hz, Hb), 6.37 (dd, 1H, J = 9.9, 17.1 Hz, Hc), 7.42 (s, 1H, NH)。
