在氮氣保護(hù)下,向3L三口燒瓶中加入濕的Degussa型Pd/C催化劑(10% Pd,50%水含量,8.120g,76.30mmol),隨后加入EtOAc(1.706L)。通過氮氣/真空循環(huán)(3次)對混合物進(jìn)行脫氣處理。然后,加入1-芐基-4-叔丁基哌啶-1,4-二羧酸酯(243.7g,763.0mmol)的EtOAc(243.7mL)溶液。在氫氣氣氛下,將反應(yīng)混合物攪拌過夜。補(bǔ)充氫氣后,繼續(xù)攪拌3.5小時。加入甲醇(60mL)以促進(jìn)沉淀物的溶解,隨后通過硅藻土墊過濾,并用甲醇洗滌。將濾液在減壓下濃縮,得到含有少量白色固體的棕色油狀物(138.6g)。通過過濾去除固體,并用最小量的EtOAc洗滌。再次將濾液減壓濃縮,得到淺棕色油狀的哌啶-4-羧酸叔丁酯(129g,收率91%)。產(chǎn)物經(jīng)1H NMR(500MHz,DMSO-d6)表征,化學(xué)位移為:δ 2.88(dt,2H),2.44(td,2H),2.23(tt,1H),1.69-1.64(m,2H),1.41-1.33(m,11H)。
