以2-(哌嗪-1-基)吡啶-5-硼酸頻那醇酯和(S)-6-溴-1-(仲丁基)-N-((4,6-二甲基-2-氧代-1,2-二氫吡啶-3-基)甲基)-3-甲基-1H-吲哚-4-甲酰胺為原料,合成(S)-1-(仲丁基)-N-((4,6-二甲基-2-氧代-1,2-二氫吡啶-3-基)甲基)-3-甲基-6-(6-(哌嗪-1-基)吡啶-3-基)-1H-吲哚-4-甲酰胺的一般步驟如下:在100 mL三頸燒瓶中加入(S)-6-溴-1-(仲丁基)-N-((4,6-二甲基-2-氧代-1,2-二氫吡啶-3-基)甲基)-3-甲基-1H-吲哚-4-甲酰胺(365 mg,0.82 mmol)、2-(哌嗪-1-基)吡啶-5-硼酸頻那醇酯(309 mg,1.07 mmol,1.3當量)、磷酸鉀(522 mg,2.46 mmol,3當量)、水以及1,4-二氧六環(huán)溶劑。隨后,在氮氣保護下加入[1,1'-雙(二苯基膦基)二茂鐵]二氯化鈀(II)二氯甲烷絡合物(53.9 mg,0.066 mmol),并將反應混合物加熱至90℃進行反應。反應完成后,經(jīng)過純化處理,得到目標產(chǎn)物400 mg,收率為92%。

在體外,EZH2野生型和突變型DLBCL細胞系中,GSK126最有效地抑制H3K27me3,其次是H3K27me2。GSK126也能有效抑制EZH2突變型DLBCL細胞系的增殖,并誘導敏感細胞系中EZH2靶基因的轉(zhuǎn)錄激活。在A687V EZH2突變細胞中,GSK126處理導致總體H3K27me3減少,強基因活化,胱天蛋白酶活化,以及增殖減少。在親代H2087細胞中,GSK126抑制VEGF-A和磷酸化Ser(473)-AK的表達,因此引起對細胞增殖,遷移和代謝的抑制。
