以化合物12(CAS: 75342-32-4,1.40g,7.7mmol)為原料,將其溶解于甲醇(30mL)中,加入過量氫氧化鉀。將反應(yīng)混合物加熱回流2小時。反應(yīng)完成后,通過旋轉(zhuǎn)蒸發(fā)儀減壓除去溶劑。將殘余物用二氯甲烷(20mL)和水(20mL)進行分配萃取。分離有機相,用無水硫酸鈉干燥,過濾后減壓濃縮,得到化合物13(5-甲氧基吡啶-2-甲醇)為淺黃色油狀物(1.00g,收率93%)。產(chǎn)物經(jīng)1H NMR(CDCl3)表征:δ 1.82(s, 1H),3.87(s, 3H),4.70(s, 2H),7.18-7.24(m, 2H),8.25(d, 1H, J = 2.8Hz);GC/MS分析顯示m/z 140(M++1, 3%),139(M+, 44%),138(100%),110(71%)。
