在配備攪拌子的2-5 mL微波反應(yīng)管中,將3-氨基丁-2-烯腈(164 mg,2 mmol)與5 mL 1 M鹽酸溶液混合,攪拌至完全溶解,制得0.4 M的原料溶液。隨后,向該溶液中加入苯肼(216 mg,2 mmol)。將反應(yīng)管用鋁蓋密封后,置于微波反應(yīng)器中,在150℃下以‘非常高’的吸光度設(shè)定照射10分鐘。反應(yīng)完成后,冷卻至室溫,得到含有橙色沉淀的非均相溶液。用10%氫氧化鈉溶液調(diào)節(jié)pH至堿性,并超聲處理5分鐘以促進(jìn)固體沉淀的形成。通過(guò)過(guò)濾收集沉淀,用去離子水洗滌兩次,干燥后獲得淺橙色固體產(chǎn)物(292 mg,收率84%)。對(duì)于不易沉淀的產(chǎn)物,可采用二氯甲烷(DCM)對(duì)堿性水層進(jìn)行三次萃取以分離產(chǎn)物。合并有機(jī)相,用無(wú)水硫酸鎂干燥,過(guò)濾后減壓濃縮,得到目標(biāo)化合物。
