4-Fluorobenzenesulfonyl Chloride is found to be an excellent activating agent for the covalent attachment of biological substances to a variety of solid supports e.g. Sepharose beads. 4-Fluorobenzenesulfonyl Chloride is also used as a reagent for the studies of proteins by fluorine NMR. | [Synthesis]
The general procedure for the synthesis of 4-fluorobenzenesulfonyl chloride from p-fluorobenzenethiol is as follows: a mixture of mercaptan (1 mmol), 30% H2O2 (3 mmol, 0.3 mL), and TiCl4 (1 mmol, 0.11 mL) was reacted in CH3CN with stirring at 25 °C for the time referred to in Table 1.The reaction was carried out by the immediate precipitation of white solid (TiO2) . The progress of the reaction was monitored by TLC, and upon completion of the reaction, the reaction mixture was quenched by the addition of H2O (10 mL) and subsequently extracted with EtOAc (4 × 5 mL). The organic phases were combined and dried with anhydrous MgSO4. The filtrate was concentrated under reduced pressure to give an analytically pure product. The product can be further purified by short silica gel column chromatography and structurally confirmed by 1H NMR, 13C NMR spectroscopic analysis and comparison of melting points with real samples prepared by known methods. | [References]
