The general procedure for the synthesis of 4-amino-3-chloro-5-fluorobenzonitrile from 3-fluoro-4-aminobenzonitrile is as follows: 3-fluoro-4-aminobenzonitrile (1 eq.) was mixed with N-chlorosuccinimide (1.5 eq.) in acetonitrile (0.24 M), and the reaction was stirred for 5 hr at 85 °C. After completion of the reaction, the solvent was removed by rotary evaporation. The residue was partitioned between ethyl acetate and 5% sodium hydroxide solution. The organic phase was washed sequentially with 5% sodium hydroxide solution and saturated saline. Subsequently, the organic phase was dried with anhydrous magnesium sulfate, filtered and concentrated under reduced pressure to give 4-amino-3-chloro-5-fluorobenzonitrile in 88% yield, the product was a beige solid.1H NMR (400MHz, CDCl3) δppm: 7.41 (t, J=6Hz, 1H), 7.24 (dd, J=1.8,10.2Hz, 1H), 4.66 (s 2H). | [References]
