成纖維細(xì)胞活化蛋白（FAP/分離酶）是一種II型跨膜絲氨酸蛋白酶，其膜結(jié)合型與可溶型均具有脯氨酸后切割內(nèi)肽酶活性，尤其偏好切割Ala/Ser-Gly-Pro-Ser/Asn/Ala共識(shí)序列；同時(shí)可降解明膠及熱變性I型膠原，并展現(xiàn)二肽基肽酶活性（優(yōu)先作用于Ala-Pro、Ile-Pro、Gly-Pro、Arg-Pro和Pro-Pro）。膜結(jié)合型FAP通過與DPP4、PLAUR或整合素結(jié)合，介導(dǎo)細(xì)胞外基質(zhì)（ECM）的細(xì)胞周蛋白水解，進(jìn)而促進(jìn)細(xì)胞粘附、遷移及侵襲；在神經(jīng)膠質(zhì)瘤中通過降解蛋白聚糖短蛋白聚糖促進(jìn)腦實(shí)質(zhì)侵襲。值得注意的是，該蛋白在黑色素細(xì)胞中通過不依賴絲氨酸蛋白酶活性的方式調(diào)控增殖與存活，發(fā)揮抑癌作用。
