轉化生長因子β1(TGFB1),亦稱TGF-β1、CED、DPD1或TGFB,是轉化生長因子β細胞因子超家族的多肽成員。該分泌蛋白具有多重細胞功能,包括調(diào)控細胞生長、增殖、分化和凋亡。TGFB1蛋白可調(diào)控細胞的生長與分裂(增殖)、細胞成熟執(zhí)行特定功能的過程(分化)、細胞運動(遷移)及細胞自我毀滅程序(凋亡)。該蛋白廣泛存在于人體各組織,在胚胎發(fā)育、血管形成、肌肉組織調(diào)節(jié)、體脂發(fā)育、傷口愈合及免疫系統(tǒng)功能中發(fā)揮關鍵作用。
TGFB1在構成骨骼的組織中尤為豐富,參與調(diào)節(jié)骨生長;同時大量存在于細胞間隙形成的復雜網(wǎng)絡結構(細胞外基質)中。在細胞內(nèi),該蛋白通常處于關閉狀態(tài)(失活),直至接收化學信號才被激活。TGFB1在免疫系統(tǒng)調(diào)控中具有重要作用,其對不同類型細胞或不同發(fā)育階段的細胞呈現(xiàn)差異化活性。大多數(shù)免疫細胞(白細胞)均可分泌TGFB1。研究證實該蛋白可與TGFβ受體1、LTBP1、YWHAE、EIF3I及核心蛋白聚糖(Decorin)發(fā)生相互作用。
