背景介紹
B淋巴細(xì)胞抗原CD19，也稱為CD19（分化簇19），是一種單次跨膜I型膜蛋白，包含兩個免疫球蛋白樣C2型結(jié)構(gòu)域（Ig-like C2-type domains）。
CD19 表達(dá)于濾泡樹突細(xì)胞和B細(xì)胞上。事實上，從發(fā)育過程中最早可識別的B系細(xì)胞到B母細(xì)胞，CD19都存在于B細(xì)胞上，但在成熟為漿細(xì)胞時丟失。
CD19 主要作為B細(xì)胞共受體（co-receptor），與CD21和CD81協(xié)同作用。被激活后，CD19的胞質(zhì)尾部會發(fā)生磷酸化，導(dǎo)致其被Src家族激酶結(jié)合，并募集PI-3激酶。
與T細(xì)胞類似，B淋巴細(xì)胞表面也有多個分子形成抗原受體復(fù)合物。（幾乎）B細(xì)胞特異性的CD19磷酸化糖蛋白就是這些分子之一。其他分子包括CD21和CD81。這些表面免疫球蛋白（sIg）相關(guān)分子有助于信號轉(zhuǎn)導(dǎo)。
在活體B細(xì)胞上，模擬外源性抗原的抗免疫球蛋白抗體會導(dǎo)致CD19與sIg結(jié)合并隨之內(nèi)化。反向過程尚未被證實，這表明該受體復(fù)合物的形成是抗原誘導(dǎo)性的。這種分子結(jié)合關(guān)系已通過化學(xué)研究得到證實。
