FAAP24 (Fanconi anemia-associated protein 24 kDa) is a DNA repair protein that plays a critical role in the Fanconi anemia (FA) pathway, a key mechanism for maintaining genomic stability. It was initially identified as a component of the FA core complex, which orchestrates the repair of DNA interstrand crosslinks (ICLs) through monoubiquitination of FANCD2 and FANCI. FAAP24 interacts directly with FANCM, another FA protein, forming a heterodimer that facilitates DNA damage recognition and stabilizes the FA core complex at stalled replication forks. Structurally, FAAP24 contains an XPA-like domain, enabling it to bind damaged or branched DNA structures, and a conserved FAAP24-specific C-terminal domain critical for protein interactions.

Antibodies targeting FAAP24 are essential tools for studying its expression, localization, and function in DNA repair processes. They are widely used in techniques like Western blotting, immunofluorescence, and co-immunoprecipitation to investigate FAAP24's role in ICL repair, replication stress response, and crosstalk with homologous recombination pathways. Dysregulation of FAAP24 is linked to Fanconi anemia, cancer predisposition, and chemotherapeutic resistance, underscoring its biomedical relevance. Research using FAAP24-specific antibodies has helped clarify its contribution to genome maintenance and potential therapeutic targeting in FA-deficient cancers.
