The KMT2A (Lysine Methyltransferase 2A) antibody is a tool used to detect the KMT2A protein, a histone methyltransferase critical for epigenetic regulation of gene expression. Formerly known as MLL (Mixed-Lineage Leukemia), KMT2A regulates transcriptional activation by catalyzing histone H3 lysine 4 (H3K4) methylation, a marker of active chromatin. The KMT2A gene is located on chromosome 11q23 and encodes a large multidomain protein containing N-terminal transcriptional activation domains (TAD), PHD zinc fingers, and a C-terminal catalytic SET domain.

KMT2A is frequently implicated in chromosomal translocations in acute leukemias, particularly infant and therapy-related leukemias, leading to oncogenic fusion proteins that disrupt normal hematopoiesis. Antibodies targeting KMT2A are widely used in research and diagnostics to identify KMT2A rearrangements or truncated isoforms, often via immunohistochemistry, Western blotting, or fluorescence in situ hybridization (FISH). They help characterize aberrant KMT2A localization or expression, aiding in leukemia subtyping and prognosis.

Additionally, KMT2A antibodies are employed in chromatin immunoprecipitation (ChIP) studies to map its genomic binding sites, elucidating its role in developmental and oncogenic pathways. Mutations or dysregulation of KMT2A are also linked to neurodevelopmental disorders, such as Wiedemann-Steiner syndrome, expanding its relevance beyond cancer biology. These antibodies are essential for unraveling KMT2A’s molecular functions and its therapeutic targeting in precision medicine.
