名稱	Apelin-13
描述	Apelin-13 is a 13-amino acid peptide sequence found in the central and peripheral nervous system that is an endogenous ligand for the G protein-coupled receptor angiotensin II protein J (APJ), with an EC50 value of 0.37 nM for APJ.Apelin-13 has vasodilatory and antihypertensive activity and can be used to study type 2 diabetic syndrome. 細胞實驗	For in vitro study, PC12 cells were used,proteases inhibitors or siRNA were used.?Protein levels were investigated by western blotting.
動物實驗	In vivo, the I/R model was induced and Apelin 13 was given intracerebroventricularly 15 min before reperfusion.?The neurobehavioral scores, infarction volumes, and some cytokines in the brain were measured.
體外活性	在培養(yǎng)的PC12細胞中，觀察到相同的保護效果。用其siRNA沉默Nrf2基因，取消了Apelin 13防止I/R引起的PC12細胞損傷、氧化應(yīng)激和炎癥的作用。AMPK的siRNA抑制削減了Apelin 13誘導(dǎo)的Nrf2表達水平，并減弱了Apelin 13的保護效果。GSK-3β與Nrf2之間的相互作用關(guān)系也通過相對過表達得到驗證。通過使用選擇性抑制劑，我們進一步確認了AMPK/GSK-3β/Nrf2上游是AR/Gα/PLC/IP3/CaMKK。
體內(nèi)活性	通過每日腹腔注射200 μg/kg的劑量連續(xù)使用4周，Apelin-13能顯著改善心臟功能、胰島素抗性以及脂質(zhì)代謝。它能顯著降低血清中TNF-α與瘦素的水平，并誘導(dǎo)在血清中Apelin-12的表達，同時明顯提高GLUT4和p-AMPKα2[2]的水平。當(dāng)通過單次劑量10和100 μM的腦室內(nèi)注射給藥時，Apelin-13能增加大部分蒼白球神經(jīng)元的自發(fā)放電[3]。
