名稱(chēng)	Triclosan
描述	Triclosan (Irgasan) is an antibacterial and antifungal agent.
細(xì)胞實(shí)驗(yàn)	CV-1 cells are maintained in DMEM supplemented with 10% FBS, seeded in 96-well plates, and transfected with an expression vector containing a specific nuclear receptor and RXR, along with a luciferase reporter containing the appropriate DNA response element. The assay is first validated with known nuclear receptor ligands as positive controls. For the CAR ligand-binding assay, CV-1 cells are transiently transfected with the expression vector containing the Gal4 DNA-binding domain fused with the ligand-binding domain of murine or human CAR and cotransfected with the luciferase reporter plasmid, mh 100-luc. The day after the transfection, the positive compound of each nuclear receptor or TCS(Triclosan) is added and the cells are incubated for an additional 24 h. The luciferase activities are measured and normalized by β-gal activity.(Only for Reference)
體外活性	在生理相關(guān)濃度下，用Triclosan處理的癌細(xì)胞顯著增加了通過(guò)腫瘤形成評(píng)估的癌細(xì)胞群體數(shù)量。Triclosan處理的細(xì)胞呈現(xiàn)出類(lèi)間充質(zhì)樣的形態(tài)，并減少了細(xì)胞間的黏附。這些細(xì)胞在降低E-鈣黏蛋白的同時(shí)，明顯上調(diào)了EMT標(biāo)志物，包括N-鈣黏蛋白、波形蛋白、蝸牛和鼻涕蟲(chóng)。Triclosan可能通過(guò)EMT過(guò)程，促進(jìn)癌細(xì)胞在分離狀態(tài)下的生存和移動(dòng)能力[2]。
體內(nèi)活性	Triclosan 在促進(jìn)組織纖維化方面發(fā)揮病理作用，導(dǎo)致肝癌發(fā)生。Triclosan不僅加快了小鼠肝細(xì)胞癌（HCC）的發(fā)展，還對(duì)肝臟損傷的長(zhǎng)期影響有所促進(jìn)。Triclosan促進(jìn)細(xì)胞纖維化和發(fā)生炎癥[1]。
