IL-12A分子背景
白細胞介素-12（IL-12）也稱為自然殺傷細胞刺激因子（NKSF）、細胞毒性淋巴細胞成熟因子（CLMF），是一種由兩個獨立基因IL-12A（p35）和IL-12B（p40）編碼的異二聚體細胞因子。IL12是由樹突細胞、巨噬細胞和人類B淋巴母細胞（NC-37）在抗原刺激下自然產生的。IL-12參與幼稚T細胞向Th0細胞的分化，在自然殺傷細胞和T淋巴細胞的活性中起著重要作用。IL-12還具有抗血管生成活性，這意味著它可以阻斷新血管的形成。白細胞介素-12亞基α（IL12A）也稱為NKSF1、CLMF1和P35，IL12A與IL-6、G-CSF顯示出顯著的序列相似性，并且僅在IL12B共表達時才發(fā)揮生物活性。IL12B缺陷小鼠對實驗性慢性炎癥性疾病的誘導具有抵抗力，而IL12A敲除小鼠則表現(xiàn)出更嚴重的形式，這表明這兩種亞基在慢性炎癥疾病的結果中具有相反的功能。
