4-Vinylphenol 存在于藥用草白花蛇舌草,野生稻中,也是葡萄酒中乳酸菌對對香豆酸和阿魏酸的代謝產(chǎn)物。4-Vinylphenol 誘導(dǎo)凋亡 (apoptosis),在體內(nèi)抑制血管形成并抑制浸潤性乳腺腫瘤生長。
4-Vinylphenol (12.5-200 μg/mL) significantly reduces cell viability in parental MDA-MB-231 cells, and the IC50 value is 109 μg/mL.
4-Vinylphenol (0.15 or 0.3 μg/mL; 3 days) reduces sphere?formation and vimentin expression on CSCs in breast cancer.
4-Vinylphenol (50, 100 μg/mL; 72 hours) significantly suppresses cell proliferation in CSC-enriched MDA-MB-231 cells. 4-Vinylphenol significantly reduces ALDH1A1 expression by 50% in CSC-enriched MDA-MB-231 cells.
4-Vinylphenol (100 μg/mL) significantly increases the expressions of caspase 3, likely sensitizing CSC-enriched MDAMB-231 cells to apoptosis.
生產(chǎn)方法
4-羥基苯乙烯的合成(微波輔助法):在100 mL錐形瓶中加入4-羥基苯甲醛(1.0 g,8.2 mmol)、丙二酸(1.69 g,16.3 mmol)和三乙胺(2-4 mL),隨后加入乙酸(10-15 mL)。錐形瓶口覆蓋松散放置的漏斗,充分搖勻后置于微波反應(yīng)器中,分批次進行微波輻射(2-8分鐘)。反應(yīng)完成后,將混合物冷卻至室溫,倒入冰水中,并用乙酸乙酯萃取。有機相依次用碳酸氫鈉溶液、稀鹽酸和飽和氯化鈉溶液洗滌,無水硫酸鈉干燥。減壓濃縮除去溶劑,得到粗產(chǎn)物。通過硅膠柱色譜法純化,洗脫劑為己烷/乙酸乙酯(9:1至6:4,v/v),得到具有甜香氣味的4-羥基苯乙烯,產(chǎn)率40%。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR和13C NMR確認:1H NMR (CDCl3) δ 7.31 (2H, d, J = 8.5 Hz, H-2和H-6), 6.81 (2H, d, J = 8.8 Hz, H-3和H-5), 6.67 (1H, dd, J = 17.8 Hz, 11.3 Hz, CH=CH2), 5.51 (1H, s, OH), 5.20 (1H, d, J = 17.8 Hz, 順式-CH=CH2), 5.14 (1H, d, J = 11.3 Hz, 反式-CH=CH2); 13C NMR (CDCl3) δ 155.5 (C-1), 136.5 (CH=CH2), 130.9 (C-4), 128.0 (C-3和C-5), 115.8 (C-2和C-6), 112.0 (CH=CH2)。
