通用方法:在攪拌條件下,將甘氨酰-甘氨酰-甘氨酸(1 mmol)緩慢加入含有鹽酸胍(15 mmol)和二碳酸二叔丁酯(2.5-3 mmol)的乙醇(1 mL)溶液中,反應(yīng)溫度維持在35-40°C。持續(xù)攪拌反應(yīng)混合物直至形成澄清溶液。隨后,在真空條件下蒸發(fā)除去乙醇。殘余物依次用蒸餾水(2 mL)和己烷或石油醚(2 mL)洗滌,以獲得幾乎純凈的Boc-甘氨酸-甘氨酸-甘氨酸。若需進(jìn)一步提高產(chǎn)物純度,可對(duì)粗產(chǎn)物進(jìn)行重結(jié)晶處理。
