向1H-咪唑(5g,73.5mmol)的四氫呋喃(THF)溶液中分批加入氫化鈉(1.8g,45mmol),室溫攪拌30分鐘。隨后,緩慢滴加溴乙腈(8.8g,73.9mmol),繼續(xù)室溫攪拌2小時。反應(yīng)完成后,通過加入水(50mL)和飽和氯化銨溶液(50mL)淬滅反應(yīng)。將反應(yīng)混合物用乙酸乙酯(100mL×3)萃取,合并有機(jī)層。有機(jī)層用無水硫酸鎂干燥,過濾后減壓濃縮。粗產(chǎn)物通過硅膠柱色譜法純化(洗脫劑:石油醚/乙酸乙酯=10:1),得到2-(1H-咪唑-1-基)乙腈(4.6g,收率59%)為黃色油狀物。LRMS m/z: 108 [M+H]+。
