以3-氟-4-溴苯基乙腈為原料合成2-(4-溴-3-氟苯基)乙酸的一般步驟:將3-氟-4-溴苯基乙腈(0.78 g,3.64 mmol)溶于硫酸(5 mL)和水(5 mL)的混合溶液中,于100℃下攪拌反應16小時。通過LCMS監(jiān)測反應進程,確認原料完全消耗后,將反應混合物溶于水(20 mL)中,并用乙酸乙酯(20 mL)萃取。合并有機層,用無水硫酸鈉干燥,過濾后減壓濃縮,得到白色固體2-(4-溴-3-氟苯基)乙酸(0.7 g,2.046 mmol,收率56.1%)。產物經1H NMR(400 MHz,CD3OD)表征:δ 7.53(t,J = 7.8 Hz,1H),7.16(dd,J = 9.8, 1.9 Hz,1H),7.02(d,J = 8.0 Hz,1H),3.61(s,2H)。
