向攪拌的4-碘吡唑(5.0 g,25.8 mmol)的二氯甲烷(10 mL)溶液中依次加入對甲苯磺酸一水合物(TsOH·H2O,0.5 g,2.9 mmol)和3,4-二氫-2H-吡喃(4.5 g,53.5 mmol)。將反應(yīng)混合物在室溫下攪拌2小時。反應(yīng)完成后,將混合物在減壓下濃縮以除去溶劑。粗產(chǎn)物通過硅膠柱色譜法純化,使用乙酸乙酯/石油醚(1:3,v/v)作為洗脫劑,得到目標(biāo)化合物4-碘-1-(四氫吡喃-2-基)-1H-吡唑(7 g,收率93%),為無色油狀物。LC-MS(ESI)分析顯示:[M-84 + H]+ = 195.1。
