以6-溴-1H-吲唑(700 mg, 3.55 mmol)和聯(lián)硼酸頻那醇酯(1.50 g, 5.91 mmol)為原料,在氮?dú)獗Wo(hù)下,將兩者與Pd(dppf)Cl2-DCM(290 mg, 0.36 mmol)和KOAc(1.04 g, 10.6 mmol)混合于DMF(20 mL)中。反應(yīng)混合物在100℃下攪拌15小時(shí)。反應(yīng)完成后,將混合物真空濃縮,殘余物懸浮于EtOAc(30 mL)中,通過硅藻土過濾,濾液濃縮后得到866 mg(產(chǎn)率100%)1H-吲唑-6-硼酸頻哪醇酯,為棕色半固體,無需進(jìn)一步純化即可用于后續(xù)反應(yīng)。LC-MS分析結(jié)果:C13H17BN2O2 + H+ [M + H]+,計(jì)算值245.1,實(shí)測(cè)值245.0。
