以(2S,5R)-5-(4-((2-氟芐基)氧基)苯基)吡咯烷-2-羧酸甲酯(6.4g,19.43mmol)為原料,將其與7M氨的甲醇溶液(52.47mL,367.27mmol)在密封燒瓶中混合并攪拌。反應(yīng)混合物在室溫下靜置4天后,通過旋轉(zhuǎn)蒸發(fā)除去溶劑。隨后加入甲苯(12mL)并再次蒸發(fā)以去除殘留的甲醇。將得到的固體懸浮于甲苯(10.5mL)中,攪拌2小時,然后加入庚烷(3.5mL),繼續(xù)在室溫下攪拌30分鐘,隨后在冰浴中冷卻并攪拌30分鐘。通過過濾收集固體,先用冷的3:1甲苯/庚烷混合溶劑(12mL)洗滌,再用庚烷(12mL)洗滌,并吸干。將固體在45℃的真空烘箱中干燥,得到米色固體產(chǎn)物(5.85g)。NMR分析顯示在芳香族區(qū)域(6.8-6.9ppm)存在微量雜質(zhì),推測可能為未反應(yīng)的苯酚。為此,將產(chǎn)物溶解于乙酸乙酯(200mL)中,依次用2% Na2CO3溶液(2×100mL)和水(100mL)洗滌,有機相用Na2SO4干燥后濃縮,得到灰白色粉末狀產(chǎn)物(5.47g,收率90%)。LC-MS分析顯示MH+ = 315(對應(yīng)分子式C18H19FN2O2)。NMR(CDCl3)數(shù)據(jù)如下:δ 1.68(1H,m),2.06-2.35(3H,m),2.51(1H,br s),3.88(1H,dd,J = 3Hz,9Hz),4.31(1H,dd,J = 6Hz,9Hz),5.15(2H,s),5.57(1H,br s),6.99(2H,d,J = 9Hz),7.11(1H,m),7.18(1H,m),7.30-7.40(3H,m),7.53(1H,m)。
