在氬氣保護(hù)下,向攪拌的3-溴-4-甲基苯甲腈(2g,10.2014mmol,純度100%)在四氯化碳(20mL,純度100%)中的溶液中,依次加入N-溴代琥珀酰亞胺(NBS,2.2g,12.2417mmol,純度99%)和過氧化苯甲酰(0.123g,0.5101mmol,純度100%)。將反應(yīng)混合物加熱至85℃,并在此溫度下攪拌反應(yīng)18小時。反應(yīng)完成后,將反應(yīng)混合物用水(50mL)稀釋,并用乙酸乙酯進(jìn)行萃取。合并有機(jī)相,依次用水和飽和食鹽水洗滌,經(jīng)無水硫酸鈉干燥后,減壓濃縮,得到3-溴-4-(溴甲基)苯甲腈(1.2g,4.4mmol,純度90%,收率95%)為灰白色固體。產(chǎn)物經(jīng)1H NMR(400MHz,CDCl3)確認(rèn):δ 3.89-4.01(m,2H),6.61-6.77(m,1H),6.92-7.07(m,2H)。LCMS分析顯示:保留時間1.12-1.49分鐘,m/z 274 [M+H]+。
