在0℃下,將N-(叔丁氧羰基)-D-纈氨酸(5g,11.5mmol)溶于THF(69mL,0.35M)中,隨后加入碘甲烷(11.45mL,8當(dāng)量)。在攪拌下,緩慢加入60% NaH礦物油分散體(2.76g,3當(dāng)量)。反應(yīng)混合物在室溫下劇烈攪拌過夜,之后用乙醚(70mL)稀釋,并在0℃下緩慢加入水(50mL)淬滅反應(yīng)。分離有機層和水層,水層用乙醚(2×30mL)萃取兩次。通過逐滴加入飽和食鹽水將水層pH調(diào)至約2(使用pH試紙檢測),隨后用乙酸乙酯(4×100mL)萃取。合并有機層,用飽和食鹽水洗滌,經(jīng)無水硫酸鎂干燥,過濾后真空濃縮。得到油狀產(chǎn)物(4.46g,收率84%),其純度足以進行下一步反應(yīng)。如需進一步純化,可通過快速柱色譜(硅膠,3%-5%甲醇/二氯甲烷)獲得分析純樣品,為白色蓬松固體。在類似條件下僅使用3當(dāng)量碘甲烷時,反應(yīng)結(jié)束時仍有原料殘留,可能是由于二鈉鹽的競爭性沉淀所致。
