以2-羥基-5-碘-3-(三氟甲基)吡啶為原料合成2-氯-5-碘-3-三氟甲基吡啶的一般步驟:在微波反應(yīng)小瓶中,將5-碘-3-三氟甲基-2-吡啶醇(1g,3.47mmol)加入預(yù)先冷卻的POCl3(1.60mL)和DMF(1mL)的混合液中。密封小瓶后,于110℃加熱反應(yīng)20分鐘。反應(yīng)完成后,將混合物冷卻至室溫,并緩慢倒入冰水中。此時(shí),產(chǎn)物以沉淀形式析出。通過(guò)過(guò)濾收集沉淀,用冷水洗滌,干燥后得到淺棕色粉末狀產(chǎn)物661mg,收率62%。產(chǎn)物的結(jié)構(gòu)通過(guò)1H NMR(500MHz, CDCl3)δ 8.32(d, J = 2.0Hz, 1H),8.81(d, J = 2.0Hz, 1H)和13C NMR(250MHz, CDCl3)δ 89.4, 121.2(q, JC-F = 273.3Hz),126.8(q, JC-F = 33.6Hz),144.34, 148.5, 158.7進(jìn)行確認(rèn)。
