以4-溴-2-碘-1-甲基苯為原料合成4-溴-1-(溴甲基)-2-碘苯(15b)的一般步驟:在氮?dú)獗Wo(hù)下,將4-溴-2-碘甲苯(3.94 g,13.3 mmol)溶于1,2-二氯乙烷(19 mL)中,置于50 mL雙頸圓底燒瓶中,室溫?cái)嚢琛kS后依次加入過氧化苯甲酰(164 mg,0.663 mmol)和N-溴代琥珀酰亞胺(NBS,2.63 g,14.6 mmol),將反應(yīng)混合物加熱回流5小時(shí)。反應(yīng)完成后,冷卻至室溫,過濾收集沉淀,并用己烷洗滌。濾液經(jīng)無(wú)水硫酸鎂(MgSO4)干燥后,過濾并減壓濃縮,得到橙色固體。該固體用冷甲醇沖洗,得到灰白色固體產(chǎn)物(3.27 g,8.75 mmol,收率66%),無(wú)需進(jìn)一步純化即可用于后續(xù)反應(yīng)。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(400 MHz,CDCl3)確認(rèn):δ 8.01(d,1H,J = 2.0 Hz),7.47(dd,1H,J = 8.3, 2.0 Hz),7.33(d,1H,J = 8.3 Hz),4.54(s,2H);高分辨質(zhì)譜(HRMS,EI)m/z計(jì)算值C7H5Br2I [M+]:377.7762,實(shí)測(cè)值:377.7759。
