以(R)-2-氯-7-乙基-8-異丙基-7,8-二氫蝶啶-6(5H)-酮(27.5 mmol, 7 g)和碳酸二甲酯(41 mmol, 5.7 g)為原料,在30 mL碳酸鉀懸浮液中反應(yīng)。將反應(yīng)混合物置于高壓釜中,加熱至130℃,維持5小時(shí)。反應(yīng)完成后,將混合物冷卻至25℃。隨后,加入25 mL去離子水和15 mL乙酸乙酯,攪拌混合。有機(jī)相通過減壓蒸餾濃縮。向殘余物中加入25 mL乙醇和45 mL去離子水,加熱至60℃溶解。將溶液冷卻至室溫,抽濾收集沉淀。沉淀用軟化水和乙醇(體積比2:1)的混合液洗滌。最后,將產(chǎn)物在50℃下真空干燥箱中干燥,得到(R)-2-氯-7-乙基-8-異丙基-5-甲基-7,8-二氫蝶啶-6(5H)-酮(6 g,產(chǎn)率82%)。
