以5-甲基-3(2H)-噠嗪酮為原料合成6-酮-1,6-二氫噠嗪-4-羧酸的一般步驟:將5-甲基-3(2H)-噠嗪酮(0.90g,8.2mmol)溶于濃硫酸(13mL)中,攪拌并加熱至45℃。在30分鐘內(nèi)分批加入高錳酸鉀(3.6g,12mmol),以控制反應(yīng)溫度避免劇烈升高。加畢,繼續(xù)于45℃攪拌反應(yīng)30分鐘。反應(yīng)完成后,冷卻至室溫,向反應(yīng)混合物中加入冰水淬滅。通過真空過濾收集析出的沉淀,依次用冷水和乙醚洗滌,得到0.98g(產(chǎn)率87%)目標(biāo)產(chǎn)物6-酮-1,6-二氫噠嗪-4-羧酸,為淺綠色固體。產(chǎn)物經(jīng)1H NMR(300MHz,CDCl3)表征:δ 13.39(寬單峰,1H),8.12(單峰,1H),7.22(單峰,1H)。
