(1a)3-溴-5-甲氧基苯酚的合成:將市售的1-溴-3,5-二甲氧基苯(18.74 g,86.3 mmol)溶于1-甲基-2-吡咯烷酮(100 mL)中,加入甲硫醇鈉(6.74 g,96.2 mmol)。在氮?dú)獗Wo(hù)下,將反應(yīng)混合物于100℃攪拌3小時(shí)。反應(yīng)完成后,將反應(yīng)液冷卻至室溫,加入1N鹽酸(200 mL)酸化,用乙醚(500 mL)萃取。有機(jī)層用飽和鹽水洗滌,無水硫酸鈉干燥。減壓濃縮除去溶劑,所得殘余物通過硅膠柱色譜純化(洗脫劑:乙酸乙酯/己烷,20%-25%梯度),得到目標(biāo)化合物3-溴-5-甲氧基苯酚(15.03 g,產(chǎn)率86%),為白色固體。1H-NMR(CDCl3, 400 MHz)δ: 3.77(3H, s), 4.82(1H, s), 6.33(1H, t, J = 2.4 Hz), 6.61(1H, t, J = 2.0 Hz), 6.66(1H, t, J = 2.0 Hz)。
