以2-氟-5-硝基芐醇(2.00 g, 11.9 mmol)為起始原料,將其與鐵粉(3.72 g, 66.8 mmol)和鹽酸(265.84 mg, 7.28 mmol)混合。將混合物在回流條件下加熱2小時(shí)。反應(yīng)完成后,趁熱通過硅藻土過濾,并冷卻至室溫。隨后,用飽和碳酸氫鈉溶液堿化水相,并用二氯甲烷(CH2Cl2)進(jìn)行萃取。分離有機(jī)層,用無水硫酸鈉(Na2SO4)干燥,過濾后,通過旋轉(zhuǎn)蒸發(fā)儀濃縮,得到棕色固體粗產(chǎn)物5-氨基-2-氟芐醇(1.51 g, 10.7 mmol, 90%收率)。產(chǎn)物經(jīng)紅外光譜(KBr)鑒定,顯示吸收峰位于3568, 3410, 2930, 1628, 1512, 1439, 1358, 1036, 874, 818, 737 cm-1;1H-NMR(CDCl3)譜圖顯示化學(xué)位移δ:7.26(寬單峰, 2H), 6.84(三重峰, J = 9.2 Hz, 1H), 6.75-6.69(多重峰, 1H), 6.61-6.50(多重峰, 1H), 4.67(單峰, 2H)。
