步驟1:3-甲基噻吩-2-羧酸甲酯的合成:將3-甲基噻吩-2-羧酸(15g,105mmol,1.0當(dāng)量)溶解于甲醇(150mL)中,緩慢滴加濃硫酸(7.5mL)。將反應(yīng)混合物加熱至回流,持續(xù)反應(yīng)12小時。反應(yīng)完成后,通過減壓蒸餾除去溶劑。將殘余物溶解于乙酸乙酯(200mL)中,依次用水(2×50mL)、飽和碳酸氫鈉水溶液(2×50mL)和鹽水(50mL)洗滌。有機相用無水硫酸鈉干燥,過濾后,減壓濃縮濾液,得到20g(產(chǎn)率97%)的3-甲基噻吩-2-羧酸甲酯,為油狀物。產(chǎn)物經(jīng)1H NMR(400MHz,CDCl3)確認:δ7.38(d,J=5.0Hz,1H),6.91(d,J=5.0Hz,1H),3.85(s,3H),2.55(s,3H);ESI-MS m/z 157([M+H]+)。
